Optimization of Preparation Conditions of Thyme Essential Oil Microcapsules by Layer Self-assembly Method

Summary

Using layer-by-layer self-assembly method, thyme essential oil emulsion nanoparticles as the core, using chitosan and sodium alginate to embed thyme essential oil; using Sipan 80 and Tween 60 compounding emulsifier hydrophilic and lipophilic balance (hydrophilic lipophilic balance (HLB) value, the amount and the ratio of oil to milk were tested by three factors to determine the optimal preparation conditions of the emulsion; the mass concentration of the wall material solution, the number of assembled layers, the assembly time of each layer, the stirring rate, the assembly temperature, The single factor test was carried out on the pH value of the wall material solution and the amount of the curing agent. The orthogonal test was designed to determine the optimal embedding conditions. The results showed that the preparation conditions of the emulsion were HLB value 10, emulsifier dosage 6%, and the volume ratio of essential oil to emulsifier 1:6. The emulsified particles prepared under this condition were template, the number of assembled layers was 4 layers, and the stirring rate was 1 040 r. /min, assembly time of each layer 15 min, wall material pH 5.0, assembly temperature 30 °C, the highest embedding rate under this condition is 80.23%.

Abstract:In this work, using thyme oil emulsion nanoparticles as the core, chitosan (CS) and sodium alginate (ALG) were employed for embedding thyme essential oil by a layer-by-layer self-assembly method. Three emulsification conditions including HLB ( Static lipophilic balance) value of 1:1 mixture of Span 80 and Tween 60, emulsifier concentration and ratio of oil to emulsifier were optimized to be 10, 6% and 1:6, respectively. The maximum microencapsulation efficiency of 80.23% was obtained when Self-assembly of the emulsion particles in four layers by alternate use of CS and ALG was conducted for 15 min at 30 °C with stirring at a speed of 1 040 r/min, as determined by orthogonal array design.
